Solid material is generally held at -20 °C or colder, while reconstituted solutions are kept at 2-8 °C and used within a short window. Buffers that maintain a slightly acidic to neutral pH tend to improve short-term peptide stability. Repeated warming and cooling of stock solutions promotes aggregation and should be avoided. Container closures should remain intact, since adsorption to some plastics can reduce the amount of peptide in solution.
Identity and purity are established with reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatographic profiles reveal related impurities, truncated sequences, and oxidation products, while mass measurement confirms the expected molecular mass. Purity values for research material are typically reported as a percentage by peak area. Reference standards help calibrate retention behavior across instruments. Independent laboratories emphasize method suitability because results depend heavily on column chemistry, gradient, and detection wavelength. Batch-to-batch comparison relies on the same validated method.

Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
Identification and purity assessment typically rely on reversed-phase high-performance liquid chromatography, often paired with mass spectrometry. Mass measurement confirms the expected molecular mass and can reveal truncations or modifications. Peptide mapping and sequencing techniques provide sequence-level confirmation when needed. Because related peptide impurities can behave similarly in a single method, orthogonal techniques are usually combined. Reported purity values depend heavily on the method used and should be interpreted with that in mind.
Dissolution behavior depends on the amino acid sequence, the counterion content, and the buffer chosen. Many peptides disperse readily in water or mild aqueous buffers, while others require a small amount of organic co-solvent or a change in pH. Adsorption to plastic and glass surfaces can reduce the concentration of a solution over time, particularly at low concentrations. Filtration before analysis removes particulates, and aliquoting limits repeated freeze-thaw cycles that stress the material.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Pharmacologically the compound activates three receptors: GLP-1, GIP, and glucagon. GLP-1 and GIP signaling contribute to glucose-dependent insulin release, delayed gastric emptying, and reduced appetite, while glucagon receptor activation is associated with increased energy expenditure and hepatic fat oxidation. The single-molecule design is intended to keep these activities in one peptide rather than combining separate agents. Relative activity at each receptor differs, and the balance between them is a central question in interpretation. The glucagon component is partly offset by incretin-mediated insulin secretion, an interaction that remains incompletely characterized.
The first ionisation energy of an element or molecule is the energy required to move the most loosely held electron from one mole of gaseous atoms of the element or molecules to form one mole of gaseous ions with electric charge +1. The factors affecting the first ionisation energy are the nuclear charge, the amount of shielding by the inner electrons and the distance from the most loosely held electron from the nucleus, which is always an outer electron in main group elements. The first two factors change the effective nuclear charge the most loosely held electron feels. Since the outermost electron of alkali metals always feels the same effective nuclear charge (+1), the only factor which affects the first ionisation energy is the distance from the outermost electron to the nucleus. Since this distance increases down the group, the outermost electron feels less attraction from the nucleus and thus the first ionisation energy decreases. This trend is broken in francium due to the relativistic stabilisation and contraction of the 7s orbital, bringing francium's valence electron closer to the nucleus than would be expected from non-relativistic calculations. This makes francium's outermost electron feel more attraction from the nucleus, increasing its first ionisation energy slightly beyond that of caesium. The second ionisation energy of the alkali metals is much higher than the first as the second-most loosely held electron is part of a fully filled electron shell and is thus difficult to remove.
Absinthe's popularity grew steadily through the 1840s, when it was given to French troops in Algeria as a malaria preventive, and the troops brought home their taste for it. Absinthe became so popular in bars, bistros, cafés, and cabarets by the 1860s that the hour of 5 pm was called l'heure verte 'the green hour'. It was favoured by all social classes, from the wealthy bourgeoisie to poor artists and ordinary working-class people. By the 1880s, mass production had caused the price to drop sharply, and the French were drinking 36 million litres (9,500,000 US gal) per year by 1910. Absinthe was exported widely from France and Switzerland and attained some degree of popularity in other countries, including Spain, the United Kingdom, the United States, and the Czech Republic. It was never banned in Spain or Portugal, and its production and consumption have never ceased. It gained a temporary popularity spike during the early 20th century, corresponding with the Art Nouveau and Modernism aesthetic movements. New Orleans has a cultural association with absinthe and is credited as the birthplace of the Sazerac, perhaps the earliest absinthe cocktail. The Old Absinthe House bar on Bourbon Street began selling absinthe in the first half of the 19th century. Its Catalan lease-holder, Cayetano Ferrer, named it the Absinthe Room in 1874 due to the drink's popularity, which was served in the Parisian style. It was frequented by Mark Twain, Oscar Wilde, Franklin Delano Roosevelt, Aleister Crowley, and Frank Sinatra.
Many coordination compounds of iron are known. A typical six-coordinate anion is hexachloroferrate(III), FeCl63−, found in the mixed salt tetrakis(methylammonium) hexachloroferrate(III) chloride. Complexes with multiple bidentate ligands have geometric isomers. For example, the trans-chlorohydridobis(bis-1,2-(diphenylphosphino)ethane)iron(II) complex is used as a starting material for compounds with the Fe(dppe)2 moiety. The ferrioxalate ion with three oxalate ligands displays helical chirality with its two non-superposable geometries labelled Λ (lambda) for the left-handed screw axis and Δ (delta) for the right-handed screw axis, in line with IUPAC conventions. Potassium ferrioxalate is used in chemical actinometry and along with its sodium salt undergoes photoreduction applied in old-style photographic processes. The dihydrate of iron(II) oxalate has a polymeric structure with co-planar oxalate ions bridging between iron centres with the water of crystallisation located forming the caps of each octahedron, as illustrated below.
== Similar conditions == Vacuoles may form in neutrophils if a blood sample is left standing for several hours prior to blood smear preparation, but this is an artifactual change with no clinical significance. Artifactual vacuoles are small and of uniform size and distribution, in contrast to toxic vacuoles whose size and placement are variable. Individuals with neutral lipid storage disease may exhibit persistent lipid-filled vacuoles in neutrophils and other granulocytes, which is a distinct phenomenon termed Jordans' anomaly.

For some individuals who experience difficulty in achieving full glanular engorgement of glans penis, they may be diagnosed with soft glans syndrome (glans insufficiency syndrome). It is often undiagnosed in the general population due to the lack of a standardized nomenclature.
==== Effects ==== RO5073012 by itself has no effect on locomotor activity in normal mice. It dose-dependently suppresses cocaine-induced hyperlocomotion (a psychostimulant-like effect) in rats, with near-complete suppression of cocaine-induced locomotor stimulation at the highest dose of RO5073012. Other TAAR1 agonists, including both partial agonists and full agonists, likewise suppress psychostimulant-induced hyperlocomotion. Conversely however, and in contrast to other TAAR1 partial agonists, RO5073012 non-significantly reduced the locomotor activity induced by amphetamine in normal mice. The reasons for this difference from other TAAR1 partial agonists are unclear, though RO5073012 has notably lower TAAR1 efficacy than other TAAR1 partial agonists. RO5073012 reduces basal locomotor activity in transgenic mice with TAAR1 overexpression. Amphetamine produces only weak locomotor stimulation in mice with TAAR1 overexpression, and RO5073012, by antagonizing the TAAR1, has been found to restore dextroamphetamine-induced hyperlocomotion in this context.
== Epidemiology == The beta form of thalassemia is particularly prevalent among Mediterranean peoples, and this geographical association is responsible for its original name. Thalassemias resulted in 25,000 deaths in 2013, down from 36,000 deaths in 1990. In Europe, the highest concentrations of the disease are found in Greece, coastal regions in Turkey (particularly the Aegean Region such as İzmir, Balıkesir, Aydın, Muğla, and Mediterranean Region such as Antalya, Adana, Mersin), in southern Spain, in parts of Italy, particularly southern Italy. With the exception of the Balearics, the major Mediterranean Islands, such as Sicily, Sardinia, Malta, Corsica, Cyprus, and Crete are heavily affected. Other Mediterranean peoples, as well as those in the vicinity of the Mediterranean, also have high rates of thalassemia, including people from North Africa and West Asia. Far from the Mediterranean, South Asians are also affected, with the world's highest concentration of carriers (16–18% of the population) in the Maldives. The disease is also found in populations living in Africa, the Americas, and in Tharu people in the Terai region of Nepal and India. It is believed to account for much lower rates of malaria illnesses and deaths, accounting for the historic ability of Tharus to survive in areas with heavy malaria infestation while others could not. Thalassemias are particularly associated with people of Mediterranean origin, Arabs (especially Palestinians and people of Palestinian descent), and Asians.
===== ADP receptor inhibitors ===== Also known as P2Y12 receptor antagonists, they work by reversibly interacting with the P2Y12 receptor to inhibit the adenosine diphosphate (ADP) receptors on platelets, thus preventing the linkage of platelets by fibrinogen. ADP-induced platelet aggregation and activation are hence hindered. Examples include clopidogrel, prasugrel and ticagrelor. Clopidogrel has a common drug interaction with CYP2C19 inhibitors, particularly omeprazole and esomeprazole which are indicated for treatment of peptic ulcer and gastro-oesophageal reflux disease (GERD). As the activation of clopidogrel requires an enzyme called CYP2C19, its inhibitors should be avoided.

The compromises contained in the agreement caused the civil war in the 26 counties in June 1922 – April 1923, in which the pro-Treaty Provisional Government defeated the anti-Treaty Republican forces. The latter were led, nominally, by Éamon de Valera, who had resigned as President of the Republic on the treaty's ratification. His resignation outraged some of his own supporters, notably Seán T. O'Kelly, the main Sinn Féin organiser. On resigning, he then sought re-election but was defeated two days later on a vote of 60–58. The pro-Treaty Arthur Griffith followed as President of the Irish Republic. Michael Collins was chosen at a meeting of the members elected to sit in the House of Commons of Southern Ireland (a body set up under the Government of Ireland Act 1920) to become Chairman of the Provisional Government of the Irish Free State in accordance with the Treaty. The general election in June gave overwhelming support for the pro-Treaty parties. W. T. Cosgrave's Crown-appointed Provisional Government effectively subsumed Griffith's republican administration with the death of both Collins and Griffith in August 1922. The day after the founding of the Irish Free State, anti-Treaty IRA men shot and killed Seán Hales an elected member of the lower house of Parliament (Dáil Éireann). The next day (8 December 1922), four imprisoned leaders of the anti-Treaty IRA (Liam Mellows, Rory O'Connor, Joe McKelvey and Dick Barrett) were executed in retaliation for the killing of Hales.
A high capacity for long-term storage of retinol means that well-nourished humans can go months on a vitamin A-deficient diet, while maintaining blood levels in the normal range. Only when the liver stores are nearly depleted will signs and symptoms of deficiency show. Retinol is reversibly converted to retinal, then irreversibly to retinoic acid, which activates hundreds of genes. Vitamin A deficiency is common in developing countries, especially in Sub-Saharan Africa and Southeast Asia. Deficiency can occur at any age but is most common in pre-school age children and pregnant women, the latter due to a need to transfer retinol to the fetus. Vitamin A deficiency is estimated to affect approximately one-third of children under the age of five around the world, resulting in hundreds of thousands of cases of blindness and deaths from childhood diseases because of immune system failure. Reversible night blindness is an early indicator of low vitamin A status. Plasma retinol is used as a biomarker to confirm vitamin A deficiency. Breast milk retinol can indicate a deficiency in nursing mothers. Neither of these measures indicates the status of liver reserves. The European Union and various countries have set recommendations for dietary intake, and upper limits for safe intake. Vitamin A toxicity also referred to as hypervitaminosis A, occurs when there is too much vitamin A accumulating in the body. Symptoms may include nervous system effects, liver abnormalities, fatigue, muscle weakness, bone and skin changes, and others.
=== Financing the war === A key element in British success was its ability to mobilise the nation's industrial and financial resources, and apply them to defeating France. Though the UK had a population of approximately 16 million against France's 30 million, the French numerical advantage was offset by British subsidies that paid for many of the Austrian and Russian soldiers, peaking at about 450,000 men in 1813. Under the Anglo–Russian agreement of 1803, Britain paid a subsidy of £1.5 million for every 100,000 Russian soldiers in the field. British national output continued to be strong, and the well-organised business sector channeled products into what the military needed. Britain used its economic power to expand the Royal Navy, doubling the number of frigates, adding 50 per cent more large ships of the line, and increasing the number of sailors from 15,000 to 133,000 in eight years after the war began in 1793. France saw its navy shrink by more than half. The smuggling of finished products into the continent undermined French efforts to weaken the British economy by cutting off markets. Subsidies to Russia and Austria kept them in the war. The British budget in 1814 reached £98 million, including £10 million for the Royal Navy, £40 million for the army, £10 million for the allies, and £38 million as interest on the national debt, which had soared to £679 million, more than double the GDP. This debt was supported by hundreds of thousands of investors and taxpayers, despite the higher taxes on land and a new income tax. The cost of the war amounted to £831 million.
Plantains have more starch and less sugar compared to regular bananas, which is why they are mostly cooked before eating. They are typically boiled or fried when eaten green, and when processed, they can be made into flour and turned into baked products such as cakes, bread and pancakes. Green plantains can also be boiled and pureed and then used as thickeners for soups. The pulp of green plantain is typically hard, with the peel often so stiff that it must be cut with a knife to be removed. Mature, yellow plantains can be peeled like typical dessert bananas; the pulp is softer than in immature, green fruit and some of the starch has been converted to sugar. They can be eaten raw, but are not as flavorful as dessert bananas, so are usually cooked. When yellow plantains are fried, they tend to caramelize, turning a golden-brown color. They can also be boiled, baked, microwaved, or grilled over charcoal, either peeled or unpeeled. Plantains are a staple food in the tropical regions of the world, ranking as the tenth most important staple food in the world. As a staple, plantains are treated in much the same way as potatoes, with a similar neutral flavor and texture when the unripe fruit is cooked by steaming, boiling, or frying. Since they fruit all year, plantains are a reliable staple food, particularly in developing countries with inadequate food storage, preservation, and transportation technologies. In Africa, plantains and bananas provide more than 25 percent of the caloric requirements for over 70 million people.
